What Is Chip-Seq?

Chromatin immunoprecipitation (ChIP) is a method that specifically enriches DNA fragments bound to target proteins in living cells to obtain information on protein-DNA interactions. The target protein was screened by ChIP, and we obtained the binding position of the protein and DNA by next-generation sequencing. ChIP-seq on the Illumina sequencing system supports almost any protein that can be immunoprecipitated by chromatin.

Customers Need to Provide

Samples

  • We recommend sending ChIP-enriched DNA at no less than 10-50 ng per sample. samples should be submitted in 1.5-1.7 mL microtubes or 2 mL screw-capped test tubes. Since DNA concentration measurements with nanodropper can be very inaccurate at low concentrations (<20 ng/µL), report the ratio of A260/280 to A260/230. We recommend that elution of DNA is best performed in PCR grade H2O, Tris, or low concentration EDTA TE (10 mM Tris, 0.1 mM EDTA).
  • For cell samples, the number of cells needs to be greater than 2 × 106.
  • For fresh/frozen tissue samples, >500 mg for animal tissues and >1 g for plant tissues; for cross-linked tissue samples, >500 mg for animal tissues and >1 g for plant tissues.

Please note that all samples need to be stored in a -80℃ refrigerator and transported on dry ice.

Please provide the specific concentration, volume, preparation time, and species origin for each sample. And inform about the specific information as well as the control and experimental samples (if there are groupings, describe in detail the grouping analysis information).
