Sanger Sequencing
Sanger sequencing has been widely used for about 50 years and is the gold standard for sequencing. Sanger method uses the principle of DNA replication, the core of which is the use of ddNTPs that lack the 3'-OH group and cannot form a phosphodiester bond with another dNTP, these ddNTPs can be used to abort the extension of the DNA strand. In addition, these ddNTPs have radioisotope or fluorescent labeling groups attached to them, and the DNA sequence of the molecule to be tested can be determined from the position of the electrophoretic band after gel electrophoresis and radiolucent autoradiography.
