What Is TCR Beta Sequencing?

TCR is located on the surface of T cells and consists of a majority of αβ TCR, and a minority of γδ TCR, which is highly diverse heterodimers. Each TCR chain consists of an antigen-recognized variable region and a constant region with three complementary decision regions (CDR). TCR α and γ chains are generated by VJ recombination, and β and δ chains undergo VDJ recombination, a rearrangement mechanism that confers diversity to the TCR library. Therefore, sequencing and analysis of the TCR library are important for enhancing the understanding of the immune system. TCR beta sequencing focuses attention on all regions of the TCR beta chain including CDR1, CDR2, and CDR3 to facilitate immuno-oncology studies.
