Full-Length Isoform Sequencing

What Is Full-Length Isoform Sequencing?
Iso-seq based on PacBio's Single Molecule, Real-Time (SMRT) technology is capable of sequencing contiguous full-length transcripts containing 5'UTR to 3'poly-A tails. This method takes advantage of the long-read sequences of triple sequencing to directly sequence full-length cDNAs of reverse transcripts without interrupting the transcript, resulting in full-length transcript isoforms. Iso-seq is widely used in medical and agricultural neighborhoods as a high-throughput method for annotating genomes, discovering novel transcripts, probing gene mechanisms, and characterizing isoform diversity.
Customers Need to Provide
Samples
- For total RNA samples, the sample content should be greater than 800 ng, the sample concentration should be greater than 60 ng/μL, the RIN value should be greater than 6.5, A260/280 = 1.8-2.2, A260/230 = 1.3-2.5, NanoDrop concentration/Qubit concentration ≤ 2.5.
Please note that the sampling or extraction process should be kept as cold as possible. All samples should be stored at -80℃, not freeze and thaw samples repeatedly, and transported on an adequate amount of dry ice.
Sample Information
Please provide the specific concentration, volume, preparation time, and species origin for each sample. And inform about the specific information as well as the control and experimental samples (if there are groupings, describe in detail the grouping analysis information).
